The 24 hour clearance test is more accurate.

Right, I understand that the 24 hr. test is more accurate, but could the difference in the 2 tests vary enough to make a difference say in 2 stages? For instance, if my creatinine was .99 on the one time test(which gave me a GFR of 64 or lower stage 2), when I do the 24 hr. test could it indicate the I am in stage 3 or 4?

It is possible. Serum creatinine can vary depending on several factors. It will even vary from lab to lab. A good way to check the accuracy is to do a BUN to creatinine ratio. This will help determine if other factors are influencing the readings.
